Minetest methods11/29/2023 ![]() When playing singleplayer, you should look at the games that are available. Now that you have it installed you can either play singleplayer, play on a local server or play online by connecting to a server. You can try to compile Minetest from the latest source on GitHub using this one-line script for installing/compiling Minetest Git, for Debian / Ubuntu / Mint If all else fails, you can build from source by cloning the engine repository and following the build instructions. Otherwise it is recommended to obtain Minetest through other means, such as the Flatpak package. If you are on a distribution with up to date enough repositories, you can install Minetest from your package manager. ![]() If you want a desktop shortcut or the like, just make create a shortcut to this executable.Inside the bin directory is the Minetest executable, minetest.exe. To run Minetest, open the extracted directory and look for the bin (binary) directory.Do NOT save to C:\Program Files\ or similar, as it will cause problems (no write access). Keep in mind you need write permissions to the folder.Extract it where you want, whether it be on your desktop, in your documents folder or in the root of your C: drive. Minetest on Windows is distributed in a portable archive.Always use this unless you are absolutely sure you're on an old machine that doesn't have a 64-bit processor. The 64-bit build is extremely recommended. ![]() The name "Minetest Game" only refers to the default game.

Cobalt price per pound today11/29/2023 Among the most widespread and conflictual claims is that it’s immensely destructive if not impossible to find enough minerals to make all the batteries that a global fleet of electric vehicles (EVs) will need. Critiques range from grid reliability to land-use, from economy to equity. Seco reserves the right to reject any or all materials that are non-conforming.A flood of recent articles, whether spontaneous or coordinated, seeks to discredit renewable energy, electric vehicles, and other elements of the climate-saving energy transition. Recyclable carbide must be free from contaminants, including ceramics, cermets and steel. ![]() The following products are not suitable for recycling: solid PCD, solid PCBN, cermets and ceramics.īoth of these materials require expensive reprocessing input, which significantly reduces any environmental benefits of recycling. WHAT CANNOT BE RECYCLED? WE CANNOT ACCEPT The following carbide products are suitable for recycling: inserts, solid drills, solid end mills, PCD/PCBN (tipped inserts only) and wear parts. WHAT CAN BE RECYCLED? USED CEMENTED CARBIDE PRODUCTS ![]() To receive recycling containers and set up your program, contact your Seco sales team member or customer service representative. The price depends on the market, and the total payment is based on the price of used carbide at the time of receipt by Seco. Upon receipt the used carbide will be sorted and weighed, and the customer will be compensated. Seco will recycle any carbide product, including bars and anvils, but no steel, ceramic or cermet. When the bin is full, the customer contacts us for collection and transportation to one of Seco’s certified recycling plants. Seco will supply you with recycling bins for you to collect your worn-out carbide tools. We strive to deliver winning solutions that encourage a safer workplace, support sustainable processes, and generate a return for the customer while safeguarding our planet for the next generation of manufacturers. Seco designed the Used Carbide Recycling Program to demonstrate our personal commitment to our customers and our shared environment. Our Recycling Program contributes to the UN’s Sustainable Development Goal 12: Sustainable Consumption and Production. By recycling, we can prolong the time before the resources are at an end, as well as reduce energy consumption by approximately 35% and carbon emissions by approximately 40%. The powder created by the recycling process (provided to Seco by certified recycling companies) has the same properties and qualities as virgin powder. Rather than rely entirely on mining virgin material, Seco reduces its impact on the environment by recycling used materials. Starbucks macchiato ingredients11/29/2023 ![]() ![]() If you really want to take this iced mocha over the top, you could also use chocolate milk, or even add in a little mint extract to make it an iced peppermint mocha. Start with an empty cup full of ice, pour your espresso shots overtop, mix in some milk and some chocolate syrup. This chocolate iced mocha is made very similarly to the vanilla latte, and you probably already have the main ingredient at home: cocoa powder! Make the simple syrup from the Vanilla recipe above, mix in a tablespoon of cocoa, and you’re good to go. Brew two espresso shots and pour them over the ice, then stir in 1 cup of milk and add vanilla syrup to your taste. Like the machiatto, start with an empty cup full of ice. But really, the vanilla latte is the easiest of all to make. Make a plain simple syrup by mixing boiling water and sugar together, then add in your flavouring of choice.įor this vanilla latte, I used plain old artificial vanilla extract but you could use the more expensive pure extract or opt for the vanilla bean if you can find them. Like the rest of the syrups here, the formula is pretty straightforward. Vanilla lattes are so easy to make and you don’t need to splurge on an expensive vanilla bean to get good flavour with this syrup. Why Starbucks only buys 100 percent arabica coffee beans Caffè Latte vs. If you don’t have dulce de leche at home (you can usually find it with the jams or in the ice cream isle of your local grocery store), simply boil a can of sweetened condensed milk for 3 hours in a large pot of gently boiling water. Brew your espresso, and pour it very delicately over the milk of your choice – I usually go with 2% but whole milk makes this drink extra creamy and over the top. Start with an empty cup full of ice, then add a couple tablespoons of the syrup. The trick to this Iced Caramel Machiatto recipe is making the rich dulce de leche caramel syrup and layering the espresso and milk it’s way easier than you think too! Of course, you can mix and match with different flavour options! Just stick to the basic formula of ice, coffee and flavoured syrup. These are three of my favourite iced coffee flavours. ![]() Choosing the right ice for your iced coffee is key! Crushed ice is the most popular kind but you can also use medium-sized iced cubes, which is what a lot of people typically have on hand, or larger blocks of ice if you plan on drinking your coffee outside (since the ice takes longer to melt).

Immune repertoire sequencing challanges11/28/2023 Camelid and fish derived sdAbs are able to bind to hidden antigens that are not accessible to whole antibodies, for example to the active sites of enzymes. Unlike whole antibodies, they do not show complement system triggered cytotoxicity because they lack an Fc region. The comparatively low molecular mass leads to a better permeability in tissues, and to a short plasma half-life since they are eliminated renally. ( right) The humanized HyHEL-10 Fv in complex with lysozyme (PDB 2EIZ) ( left) The nurse shark V NAR single domain in complex with lysozyme (PDB 1T6V). The complex of a single domain antibody and a protein antigen reveals a buried binding site. Some species have been shown to be active in the intestine after oral application, but their low absorption from the gut impedes the development of systemically active orally administered single-domain antibodies. Stability towards gastric acid and proteases depends on the amino acid sequence. In contrast to common antibodies, two out of six single-domain antibodies survived a temperature of 90 ☌ (194 ☏) without losing their ability to bind antigens in a 1999 study. Those derived from camelid and fish antibodies are less lipophilic and more soluble in water, owing to their complementarity-determining region 3 (CDR3), which forms an extended loop (coloured orange in the ribbon diagram above) covering the lipophilic site that normally binds to a light chain. These peptides have similar affinity to antigens as whole antibodies, but are more heat-resistant and stable towards detergents and high concentrations of urea. Properties Ī single-domain antibody is a peptide chain of about 110 amino acids long, comprising one variable domain (V H) of a heavy-chain antibody, or of a common IgG. ![]() Single-domain antibodies are being researched for multiple pharmaceutical applications, and have potential for use in the treatment of acute coronary syndrome, cancer, Alzheimer's disease, and Covid-19. ![]() The smaller size and single domain make these antibodies easier to transform into bacterial cells for bulk production, making them ideal for research purposes. They are easily isolated using the same phage panning procedure used for antibodies, allowing them to be cultured in vitro in large concentrations. Ĭamelid NANOBODIES® have been shown to be just as specific as antibodies, and in some cases they are more robust. Although most research into single-domain antibodies is currently based on heavy chain variable domains, NANOBODIES® derived from light chains have also been shown to bind specifically to target epitopes. An alternative approach is to split the dimeric variable domains from common immunoglobulin G (IgG) from humans or mice into monomers. Cartilaginous fishes also have heavy-chain antibodies (IgNAR, 'immunoglobulin new antigen receptor'), from which single-domain antibodies called V NAR fragments can be obtained. The first single-domain antibodies were engineered from heavy-chain antibodies found in camelids these are called V HH fragments. With a molecular weight of only 12–15 kDa, single-domain antibodies are much smaller than common antibodies (150–160 kDa) which are composed of two heavy protein chains and two light chains, and even smaller than Fab fragments (~50 kDa, one light chain and half a heavy chain) and single-chain variable fragments (~25 kDa, two variable domains, one from a light and one from a heavy chain). Like a whole antibody, it is able to bind selectively to a specific antigen. The extended CDR3 loop is coloured orange.Ī single-domain antibody ( sdAb), also known as a NANOBODY®, is an antibody fragment consisting of a single monomeric variable antibody domain. ![]() ![]() China.Antibody fragment Ribbon diagram of a llama V HH domain. 1974 winnebago brave11/28/2023 ![]() The first couple of hours, bouncing along at 50mph in the right lane, were absolutely terrifying. My plan was to drive in daylight and sleep in motels because nothing had been done yet to the interior or any of the camping equipment/systems. And, just to be clear, the Winnebago was still really gross. I also bought a pen so I could keep a log of how much gas I was going through (a lot). My first stop was a Wal-Mart where I picked up a decent GPS I could stick on the dash that would give me a rough idea of how fast I was going and how many miles I’d traveled. This journey was made even more interesting by the fact the speedometer, odometer, and gas gauge were still broken. Not running out of gas would get tricky but I was pretty sure I wouldn’t be speeding. ![]() I wasn’t sure how much of that VW technical experience would carry over to the Winnebago, but I knew how to expect the unexpected and keep on rolling. Without knowing much about engines or cars in general I was always having to figure stuff out with the bus. I had gotten used to this feeling when we had our ’77 bus. I pretty much knew SOMETHING would go wrong, just not exactly what or how bad it would be. Also, in general, it is not ideal to go on a 1000 mile/three day trip in a vehicle that has been sitting so long. SO after much discussion with the mechanic, I decided just to drive it like that and get it looked at in Indianapolis-if I made it. The mechanic who’d been working on it wasn’t set up to do it and none of the truck places could fit me in. ![]() I needed to get an alignment and to have the suspension looked at, but I didn’t have time. And I kind of felt like I was going to fall out of my seat a lot of the time. Basically I had to steer at an angle to go straight. But it was leaning pretty heavily to the right and the steering wheel was not aligned. When I finally went back down to pick it up it was running. It stayed at the shop for like six weeks.
